Internationally recognized competencies in the fields of structure-function relationships, analysis of transcription and its control have been developed in IGBMC. Important contributions related to nuclear receptors (NR) and basal transcription factors were obtained in this field. Integration has brought together biological and structural approaches to target important medically relevant molecules: this can be illustrated by the first determination of structures from several important NR binding domains (RXR, RAR, VDR), a functional and structural classification of NRs and the determination of numerous NRs ligand-binding domains in complex with agonists or antagonist molecules and co-activator fragments. The role of ligands and NR phosphorylation in the control of the regulation process represents an important research interest of the group.
IGBMC actively provides developmental expertise in target identification, soluble protein production including expression and characterization and a synergy of techniques for structure determination from low to high resolution including SAXS, cryo-electron microscopy, NMR, and X-ray diffraction. The implementation of new strategies for protein co-expression, the development of original methodologies for the characterization of the targets in their liganded state by native mass spectrometry, automated cloning and crystallization are key strengths of this partner.
